چکیده

Aim: Mitochondrial DNA (mtDNA) polymorphisms can be considered as a molecular marker in susceptibility to various types of cancer. In this study, we aimed investigate the potential relationship mtDNA with disease etiopathogenesis patients hematological malignancy.
Material and Methods: This study was carried out participation 80 diagnosed malignancy healthy individuals Department Medical Genetics, Atatürk University. all participants, 13 polymorphism regions 6 coding genes were investigated by Polymerase chain reaction restriction fragment length (PCR-RFLP) method. SNPs evaluated study; NADH dehydrogenase subunit 5-13704 (C/T), Cytochrome b 15315 12S rRNA 740 (G/A) 680 (T/C), C Oxidase I 7319 -7444 (G/A), II 8252 (C/G), 7660 7975 (A/G), 8014 8113 (C/A), 8152 tRNA lysine 8310 (T/C) identified as.
Results: ND-5 13704 (C>T) statistically significant malignancies compared controls (p = 0.001). There no difference between other polymorphisms.
Conclusion: Although findings obtained from suggest that may play role malignancies, large-scale studies are needed determine importance polymorphic region.

Investigation of Polymorphisms in Non-Coding Region of Human Mitochondrial DNA in 31 Iranian Hypertrophic Cardiomyopathy (HCM) Patients

The D-loop region is a hot spot for mitochondrial DNA (mtDNA) alterations, containing two hypervariable segments, HVS-I and HVS-II. In order to identify polymorphic sites and potential genetic background accounting for Hypertrophic CardioMyopathy (HCM) disease, the complete non-coding region of mtDNA from 31 unrelated HCM patients and 45 normal controls were sequenced. The sequences were aligne...

Inosine Triphosphate Pyrophosphohydrolase (ITPA) polymorphic sequence variants in adult hematological malignancy patients and possible association with mitochondrial DNA defects

BACKGROUND Inosine triphosphate pyrophosphohydrolase (ITPase) is a 'house-cleaning' enzyme that degrades non-canonical ('rogue') nucleotides. Complete deficiency is fatal in knockout mice, but a mutant polymorphism resulting in low enzyme activity with an accumulation of ITP and other non-canonical nucleotides, appears benign in humans. We hypothesised that reduced ITPase activity may cause acq...

Multidrug resistance in hematological malignancy.

The recent treatment of hematological malignancies appears to be unsatisfactory in child and adult patients with acute myeloid leukemia and adult patients with acute lymphocytic leukemia. A major problem in the treatment of leukemia is caused by the development of drug resistance to chemotherapeutic agents, which is already present at diagnosis or after chemotherapy as a minimal residual diseas...
